Responses of Rhodococcus strains to heavy metal stress
Volf, Jan ; Štěpánek, Václav (advisor) ; Martínková, Ludmila (referee)
Increasing heavy metal pollution can be dealt with not only through physical and chemical decontamination processes but also utilizing microbial bioremediation, a much more environmentally friendly way. Members of genus Rhodococcus are suitable candidates for effective bioremediations of heavy metals due to their considerable adaptability and extreme resistance to different stress conditions, including those related to presence of heavy metals or their toxic compounds. Individual rhodococcal strains are more resistant to heavy metals than most of other microorganisms and they are capable of biotransformating them to less toxic forms or at least of accumulating effectively or adsorbing them on produced polysaccharides and specific surface active substances. Their bioremediation potential is very high even though, so far, only molecular mechanisms of rhodococcal resistances to toxic arsenic compounds have been studied more in detail.
The stress response to cardiac catheterisation. The concentration of stress markers of the hypothalamic-pituitary-adrenal axis
Skarlandtová, Hana ; Kittnar, Otomar (advisor) ; Gregor, Pavel (referee) ; Svoboda, Miroslav (referee)
In our study, we analyzed four stress markers (cortisol, cortisone, DHEA and DHEAS) in blood serum in young sows using minimally invasive heart catheterisation as the stress factor. The marker levels were assessed in four defined periods of the experiment, beginning with the baseline level on the day before intervention (1), the second period was after the introduction of anaesthesia (2), the third was after conducting tissue stimulation or ablation (3), and the final period was after the end of the catheterisation (4). Cortisol and cortisone were detected using HPLC method, DHEA(S) by commercial kits. For statistical analyses non-parametric tests were used (due to non standard Gaussian data distribution). In our study we arranged these experiments: 1. Diurnal variability in these markers concentration during heart catheterisation was tested. 2. Are there differences between stress markers concentration the day before experiment (sampling 1) and in the day of the catheterisation (samplings 2, 3, 4)? 3. Are there differences between these markers concentration during operation (the first sampling was excluded)? 4. The cortisol/cortisone ratio was calculated. We found only minimal statistical differences in studied markers between the morning and afternoon group (p > 0.05) in experiment 1. For tested...
Možnosti hodnocení vitality u senescentních stromů
Ovesná, Jana
Visual assessment is nowadays the most common method of evaluation of species and determination the level of vitality of the tree is its integral part. This work is aimed at clarifying the aging process and terms of vitality and senescence. It is also intended application of available methods of evaluating the vitality of the trees in the senescent ontogenetic stage, finding the strengths and weaknesses of these procedures and propose a new method which is suitable for application to senes-cent trees. Suitable swatches on this new method is verified and compared with results of conventional methods of evaluation vitality of trees.
Stress and functional disorders of stomach
Marková, Jana ; Maruna, Pavel (advisor) ; Vokurka, Martin (referee)
This bachelor thesis deals with issues of stress and relationship between stress and functional disorders of the stomach. Theoretical part describes stomach - its anatomy and physiology with emphasis on regulatory mechanisms. In the theoretical part, stress is handled complexly - from the physiological, pathophysiological and psychological point of view. The stress response is described in detail with its effects on the human organism. Separate chapters are dedicated to the effects of chronic stress on the human psyche and gastrointestinal tract. Functional disorders of the stomach are precisely explored - its classification, epidemiology, pathophysiology, diagnostics and therapy are described. Practical part has two parts. First part is an online anonymous questionnaire study distributed among the general public. This questionnaire deals especially with the stress - frequency of feelings of stress, symptoms of stress and how can stress affect the health of the research sample. This part also studies awareness of functional gastrointestinal disorders in general public. Second study examines problematics of functional disorders of the stomach amongst patients of IV. internal clinic of Všeobecná fakultní nemocnice. It is designed as a broad questionnaire study focused on circumstances of the...
Cell cycle of astaxanthin producing green alga Haematococcus pluvialis, culture media and cultivation conditions
Vávrová, Karolína ; Němcová, Yvonne (advisor) ; Pichrtová, Martina (referee)
Haematococcus pluvialis is unicellular green alga (Chlorophyceae, Chlamydomonadales), which can be found in ephemeral rock pools and birdbaths. It is the best natural producer of strong antioxidant astaxanthin, red secondary carotenoid used as coloring agent in aquaculture and poultry breeding. Astaxanthin also has various positive effects on human health. Haematococcus pluvialis has quite complex life cycle consisting of four life stages, biflagellated zoospores, non- motile round palmella stage, thick-walled akinetes (aplanospores, cysts) with high content of astaxanthin and small biflagellated gametes. Akinetes, which enable H. pluvialis to survive desiccation in its natural habitat, are formed in response to stress conditions such as high irradiance and temperature and nutrient depletion. Cells undergo dramatic ultrastructural changes during aplanospore formation. Chloroplast volume is decreased, synthesis of high amount of astaxanthin and fatty acids results in presence of lipid droplets that spread from the center to the periphery of the cell and thick cell wall containing algaenans is formed. Cultivation of H. pluvialis is often performed in two subsequent stages, green stage focused on biomass production and red stage focused on astaxanthin accumulation under stress conditions. There are...

Stress and its impact on a performance at work
Žroutová, Martina ; Michálek, Pavel (advisor) ; Josek, Mikuláš (referee)
The bachelor thesis is about current stress problems focusing on employees in the medical establishment. The thesis is dividend into two major parts, theoretical and practical. I will read and review specialized literature about stres, stress reaction, stressors, stress measure, burnout measure and others. The empirical part is dividend into two parts. The first one is a questionnaire survey. It will determine if there is a diference in perception of stress in diverse age categories, if absence of exercise has an effect on stress or if women with children are more susceptible to stress. In the second part is used Meister questionnaire, which compares perception of stress between age categories.
A holistic approach to the bereaved in case of a sudden death from a nurse´s point of view a member of the medical emergency services
GUTOVÁ, Alena
The dissertation deals with holistic approach of nursing staff of the Rescue Service by providing pszchosicial support and crisis communication to the survivors or direct sudden death participants from nurse´s }member of the Rescue Service team]perspective. The goal of the dissertation has been 1. To detect problems which nursing staff members of the Rescue Service are prepared for communication with the survivors.
